TheAnalyticCrystallizerkitcomprisesatwo-stepmethodforoptimalcrystallizationscreeningofdetergent-solubilizedintegralmembraneproteins(IMPs)inamannerindependentofdetergentidentity.Thekitincludesasolubilityprescreen(Optimizer)thatisusedtodeterminetheappropriateproteinconcentrationforusewiththe96-conditioncrystallizationscreen(Crystallizer).TheuseofOptimizerallowstheexperimentertoreducetheamountofcrystallizationconditionstobescreenedinthefirstscreeningpassandtoincreasethelikelihoodofcrystallizationsuccess.
TheconditionsforCrystallizerhavebeenformulatedtoavoidredundancyandmaximizecoverageofcrystallizationspace.PrecipitantsandsaltswereminedfromtheconditionsmostfrequentlyusedinsuccessfulmembraneproteincrystallizationexperimentsasavailableintheMembraneProteinDataBank.AlsosampledaresixbuffersrangingfrompH4.5to9.5and18distinctsalts.
Theformulationsprovidedherehavebeenfurthertunedtorenderthescreendetergent-independent,meaningthattheprecipitantandsaltconcentrationshavebeenselectedtominimizephaseseparationthattypicallyoccursforsolutionscontaininghighconcentrationsofdetergents.
