Selectorisarapid,highthroughputassaythatexploresdetergentspacetoidentifythosebestsuitedtostABIlizethemembraneproteinofinterestinadvanceofdownstreamproteincrystallizationtrials.Theassayusesdifferentialfiltrationin94differentdetergentstogenerateinformationregardingboththestabilityandthesizeoftheprotein-detergentcomplex.Thestepsofthisassayaresimple:First,theproteinisboundtoanaffinityresin,washedandelutedinanewdetergent.Theeluateisthenpassedthroughtwofilterplateswithdifferingmolecularweightcut-offs(MWCO).Thefiltrationprofilesprovidestabilityandrelativesizinginformationthatallowstheselectionofdetergentsmostlikelytostabilizeaproteininamannerthatwouldbecommensuratewithcrystallizationandstructuredetermination.
WhyshouldIexploresuchavarietyofdetergents?IntegralmembraneproteinspresentauniquechallengetoproteinchemistsandstructuralBIOLOGists.Intheirnaturalenvironment,theseproteinsexistwithinamosaiclipidbilayerthatisadynamicandcomplexenvironment.Priortostructuralstudy,however,thesemustbeextractedfromthecellmembraneandsolubilizedinsuchawayastosatisfythehydrophobicnatureofthetransmembraneregionswhileensuringthathydrophilicdomainsareincontactwithanaqueousphase.Whileasignificantnumberofdetergentsmaybecapableofkeepingtheproteininasolubleform,thestability,homogeneityandactivityofanygivenmembraneproteincanbetremendouslyaffectedbythepropertiesofthesolubilizingcomponent.
DepositstotheProteinDataBankformembraneproteinscomprisebroadrangeofdetergentanddetergent-lipidcomplexesaspartoftheproteinpreparationsleADIngthediffraction-qualitycrystals.Thus,whilecertaindetergents(DM,DDMorOG,forexample)areknownfortheirsuccessinstabilizingintegralmembraneproteinsforstructuralstudiesandarereportedathighfrequency,significanteffortwouldberequiredtoexploredetergentspace,eventocoverdetergentspreviouslysuccessfulforproteincrystallization.
